Effect of Electric Stimulation on the Cardiac Function in Isolated Rabbit Heart

Abstract
Electrical Stimulation from maintaining of cardiac rhythmicity has been widely used in isolated Langendorff perfused hears. However, functional and biochemical differences between electrically paced and spontaneously beating hearts are not well documented. In the present study, cardiac function, creatine kinase(CK) leakage, contractile apparatus between these hearts were compared. Hearts isolated from new Zealand White rabbits (1.5-2.0 kg of body weight) were retrogradely perfused by Langendorff technique with electrical stimulation (150/min, 4-volt, 0.5 msec interval;electrically paced heart, n=7) or not (spontaneously beating heart, n=7). In comparison with the spontaneously beating hearts, left ventricular developed pressure, rate-pressure product, contractility, left ventricular end-diastolic pressure, and coronary flow rate were tended to be constantly maintained in the eletrically paced hearts, however, any significant differences were found between study groups. In addition, there were no significant differences in the creatine kinase leakage and electrophoresis pattern of the contractile apparatus between study groups. These results indicate that the electrical stimulation may not be a sine qua non condition in experiments with isolated perfused hearts. However, it may be emphasized that perfusion pressure or perfusion rate as well as electrical stimulation should be properly controlled according to species of experimental animal and body weight to get more reliable and less erroneous results since cardiac function especially the contractility could be influenced by perfusion pressure, perfusion rate, and/or oxygen or calcium concentration in the perfusate.
